Grapefruit contains furanocoumarins, natural compounds that interfere with enzymes and transporters responsible for metabolizing certain drugs in the small intestine. One of the most affected enzymes is cytochrome P450 3A4 (CYP3A4), which plays a key role in breaking down many medications.
When grapefruit blocks CYP3A4, medications that rely on this enzyme stay in the body longer than intended, leading to higher drug concentrations in the bloodstream. This can intensify side effects or even lead to toxicity. On the other hand, grapefruit can also reduce the absorption of some drugs, making them less effective.
